化學性質：奧利司他是長效和強效的特異性胃腸道脂肪酶抑制劑，常溫下為白色或類白色粉未，不溶于水，溶于氯仿，易溶于乙醇，通過與胃和小腸腔內胃脂肪酶和胰脂肪酶的活性絲氨酸部位形成共價鍵使酶失活。

用途：臨床上可應用于肥胖癥及高脂血癥。一般情況下，一次可口服120mg，一日三次，于餐中或餐后1小時服用。服藥2周后體重可開始下降。可連續服用6~12個月，如劑量增大至每日400mg以上時，其作用不再增強。

不良反應使用奧利司他已有罕見的轉氨酶升高、堿性磷酸酶升高和重度肝炎的報告，并出現肝衰竭病例，其中部分患者需要進行肝移植或可直接導致死亡。奧利司他還有發生罕見過敏反應的報道，主要臨床表現為瘙癢、皮疹、蕁麻疹、血管神經性水腫、支氣管痙攣和過敏性反應，出現大皰疹十分罕見。上市后監測還發現有*炎的報道。

Chemical properties: orlistat is a long-acting and potent specific gastrointestinal lipase inhibitor, white or white powder at room temperature, insoluble in water, soluble in chloroform, soluble in ethanol, and inactivated by covalent bonding with the active serine sites of gastric lipase and pancreatic lipase in the stomach and small intestine.

Application: it can be used for obesity and hyperlipidemia. In general, 120mg can be taken orally at one time, three times a day, during meals or one hour after meals. Weight loss may begin after 2 weeks of medication. It can be used continuously for 6 to 12 months. If the dose is increased to more than 400mg per day, its effect will no longer be enhanced.

Adverse reactions to the use of orlistat have been reported with rare elevated transaminases, elevated alkaline phosphatases, and severe hepatitis, as well as cases of liver failure, some of which require liver transplantation or may lead directly to death. Orlistat also has the occurrence rare allergic reaction report, the main clinical manifestation is pruritus, rash, urticaria, angioneurotic oedema, bronchospasm and allergic reaction, the occurrence of large herpes is very rare.
